CDI Laboratories, the NextGen Proteomics™ company born from a result of intensive efforts and advances made by synthetic biologists at the Johns Hopkins University School of Medicine.

The company that was created by scientists for scientists to address critical needs required accelerate research, advance discoveries, and translate these discoveries into novel products and services that improve human health.

 

KEY SERVICES

 

1. HuProtTM Human Proteome Array Service

The world’s largest human protein library — on a slide. Contains >21,000 full-length human proteins and is the most powerful tool ever created for mapping antigen-specific immunity.

  • Analyse antibodies in fluid or more.

(i.e. peripheral serum/plasma, cerebrospinal fluid, tissue lysates, stool, saliva), or in a synthesized sample, for any isotype (i.e. IgM, IgG, IgA, IgD, IgE). Secondary antibody binding is then quantified using fluorescence intensity measurements of features on the array. Obtain RAW readouts – and optionally – normalized readouts and group-comparison statistics.

  • More than just serum profiling.

Other profiling includes nucleotide binding, antibody specificity, small molecule binding, protein-protein interactions, and more. Dozens of academic papers have been published on HuProt, demonstrating the importance of antigen-specific immunity for everything from Alzheimers to lupus to cancer. CDI have even helped predict side-effects to immuno-oncology drugs prior to treatment.

HuProt™ service sample requirements:

Human serum or plasma: Send two 10μL aliquots per sample.

Antibody isolates: At least 3μg per sample (conc. >0.1mg / mL).

Human (CSF) cerebrospinal fluid: At least 1.5mL per sample.

Protein / peptide / small molecule: At least 3μg per sample.

DNA / RNA: At least 300μL per sample (conc. >2μM).

Cohort balancing (studies >48 samples):  Individual samples are run on individual HuProt slides; these are produced in batches of 600 and our lab team typically processes x48 samples per day.  While data from multiple days and print batches is easily pooled by our informatics pipeline, customers running larger studies may wish to consider balancing case/control and replicate samples within and across both processing days and print batches.

2. HuScan™: PhIP-Seq Antibody Profiling

Human proteome epitope-level autoantibody profiling service via T7 phage display and immunoprecipitation sequencing. Analyse patient samples (serum, plasma, CSF, etc.) for the presence of autoantibodies against the full UniProt human proteome in a single-well assay with quantitative sequencing readouts.

  • CDI Antygen™ HuScan™ detects antibodies against 29,371 unique human proteins.

HuScan™ service sample requirements:

Human serum or plasma: Send two 10μL aliquots per sample.

Human (CSF) cerebrospinal fluid: Send two 100μL aliquots per sample.

Ten sample minimum.

Cohort balancing (studies > 88 samples):  Individual PhIP-Seq studies are prepared in x96 well plates using aliquots of our phage library; each study requires x8 protein A/G beads-only internal control samples per sequencer run (provided free-of-charge).  Data are most reproducible within a single sequencer run. Sequencer runs are currently limited to x3 96-well plates (288 experimental samples + x8 protein A/G beads per run).  While data from multiple days is easily pooled by our informatics pipeline, customers running larger studies may wish to consider balancing case/control or replicate samples within and across processing days.

3. VirScanTM Viral PhIP-Seq Service

Service provides an unparalleled way to assay a patient’s viral history by comprehensively analyzing antiviral antibodies to a phage library displaying peptides from >1300 strains of viruses — nearly every virus annotated to have human tropism in the UniProt database. This product is available as a service to both academic and industry customers.

Understanding Your HuScan and VirScan Results

VirScan™ service sample requirements:

Human serum or plasma: Send two 10μL aliquots per sample.

Human (CSF) cerebrospinal fluid: Send two 100μL aliquots per sample.
